Hij is bekend van zijn eigen sitcom Malcolm & Eddie, welke hij van 1996 tot 2000 samen met zijn vriend Malcolm-Jamal Warner maakte. Hij speelde verder in films als Deuce Bigalow: European Gigolo en Scary Movie 3.
Eddie Griffin heeft vanaf begin jaren negentig een carrière opgebouwd waar vele comedy-collega’s alleen maar van kunnen dromen. Dankzij optredens als ‘regular’ in de Comedy Store – waarbij hij al snel werd vergeleken met gerenommeerde comedians als Richard Pryor, Redd Foxx en Lenny Bruce- werd hij gevraagd om opener te zijn van de toen immens populaire Andrew Dice Clay tijdens een show in Madison Square Garden.
